Mechanisms by which genetic variant in the TCF7L2 gene causes diabetes. We aim to describe the molecular mechanisms where by variants in the TCF7L2 gene increases the risk of developing type 2 diabetes. This will be achieved by investing the functional effects of differential splicing of TCF7L2 in a model system as well as in human pancreatic islets. i.e. effects on hormone secretion, target gene transcription and beta-cell proliferation. Novel transcriptional networks regulated by TCF7L2 will also be investigated.
